Fluo-Break demulsifier is a transparent fluoride chemical solution specially formulated to break down emulsion droplet microspheres, thereby readily recover internal (the dispersed phase) product.
Fluo-Break effectively break down stable emulsion droplets formed using FluoSurf surfactant, Surf droplet oil, or PFPE-PEG-PFPE surfactant. This facilitates user to recovery droplet phase/dispersed phase materials such as cells, bacteria, nanoparticles, proteins, and vesicles for subsequent analysis and characterization.

How to use?
- Transfer the emulsion droplets into EP centrifuge tubes. The upper layer is the emulsion droplet, while the lower layer is the oil phase containing surfactants such as Fluo-Oil 7500, Fluo-Oil 135, Fluo-Oil 200, or Fluo-Oil 40 fluorinated oil.
- Gently and carefully remove the excess oil phase liquid (lower layer solution) using the tip of a pipette.
- Add Fluo-Break emulsion breaker (1x the volume of the droplet microsphere emulsion) to the EP centrifuge tube and cap it*.
- Gently shake the EP centrifuge tube by hand, and the droplet microspheres will begin to rupture.
- Place the EP centrifuge tube in a microcentrifuge and centrifuge for 1 minute. The droplet microspheres will be completely disrupted, achieving two-phase separation. The upper layer is the aqueous phase, while the lower layer is a mixture of the oil phase and the demulsifier.
- Using the pipette tip, remove the upper layer of clear liquid and transfer it to a clean Eppendorf tube. This clear liquid can be used for further experiments.
For emulsion droplets stabilized with high-concentration surfactants, we suggest you to increase the double volume of Fluo-Break.
